Molecular Detection and Typing of Mycobacterium avium Subspecies paratuberculosis from Fecal Samples of Small Ruminants

The chronic nature of Jhone’s disease and prolonged incubation period the agent often complicate diagnosis this disease. present study was undertaken with an aim to compare fecal microscopy using Ziehl-Nielsen (ZN) staining method Polymerase Chain Reaction assay for detection Mycobacterium avium subspecies paratuberculosis (MAP) in small ruminants. A total 205 samples sheep goat were first microscopically examined ZN later tested presence MAP DNA through IS900 PCR. Win Episcope 2.0 software used determine agreement between test assays. Of samples, 78 (38%) positive by 48 (23.4%) Moderate level (0.517) observed Restriction enzyme analysis amplicons from IS1311 PCR revealed that all belonged Bison type strain. Single predominant strain (Bison type) among ruminants indicates its ability infect multiple host species, possibility interspecies transmission endemic co-circulation.
